Why Settle for Less?

According to the Centers for Disease Control and Prevention (CDC), over 100.1 million people in the United States are affected by obesity. This condition is associated with most chronic diseases and is the leading cause of heart disease, stroke, diabetes, and even premature death. Additionally, obesity is linked to an increased risk of certain cancers. Even modest weight loss—around 5% to 10% of your total body weight—can yield significant health benefits, including improvements in mood, blood pressure, cholesterol levels, and blood sugar.

Peptides: What is Semaglutide?

Semaglutide is a peptide that mimics the natural hormone glucagon-like peptide-1 (GLP-1), which is secreted by nutrient-responsive enteroendocrine cells (L cells) in the intestine. GLP-1 is a key incretin hormone involved in regulating glucose levels in the body.

By enhancing insulin secretion and lowering blood sugar levels, Semaglutide also helps reduce appetite and cravings. This peptide offers a valuable treatment option for adults with excessive body fat or a body mass index (BMI) over 30, making it an effective addition to a comprehensive weight management program.

How does it work?

GLP-1 plays an important role in digestion by delaying gastric emptying and suppresses appetite centers in the hypothalamus. GLP-1 plays a vital role in improving insulin resistance and production of VLDL (Very Low-Density Lipoprotein) via an intrinsic gut-liver signaling pathway.

Peptides are short chain amino acids connected with a peptide bond. Peptides promote weight loss by promoting food satiety and increasing muscle mass. The GLP-1 RA drug class is associated with multiple benefits; they have a well-documented safety profile, reduce body weight, improve blood pressure, lipid profile and other cardiovascular risk factors as well as glucose metabolism.

Since Semaglutide is a glucagon-like peptide-1 (GLP-1) analogue, it is classified as a GLP-1 receptor agonist (GLP-1 RA). It has a 94% homology to human GLP-1 and a long half-life suitable for once-weekly dosing. They are available from prosperIV through weekly subcutaneous injections administered by one of our professional registered nurses by being a part of one of our Monthly club membership wellness programs.

What to expect:

Since Semaglutide affects how your body processes and digests food, it may initially cause gastrointestinal symptoms such as nausea, vomiting, stomach upset, diarrhea, tiredness, dizziness, and constipation. These symptoms typically lessen as your body adjusts to the medication. If any of these effects persist for more than three days or worsen, contact your provider, PCP, or pharmacist immediately.

To maximize the benefits of Semaglutide and support your weight management efforts, it’s crucial to adopt a healthy lifestyle. This includes:

  • Eating Nutritious Foods: Focus on a balanced diet rich in plant-based and organic foods. Incorporate macronutrients such as proteins (e.g., seafood, lean meats, eggs, beans, nuts), complex carbohydrates (e.g., fruits, vegetables, whole grains), and healthy fats to help curb cravings.
  • Regular Exercise: Engage in at least 30 minutes of physical activity, 3-4 times per week, to help burn calories and maintain an active metabolism.

Most individuals start to notice results within 8 to 12 weeks of starting Semaglutide treatment. Clinical trials indicate that participants can achieve an average weight reduction of 5-10% with weekly doses. It’s important to proceed gradually to avoid rapid weight loss and potential side effects.

*Patients should expect to commit for 3 months, each month is evaluated by the medical director before next steps are recommended.

What is the clinical data that supports this?

Clinical Trials for Anti-obesity Effect of Semaglutide

Semaglutide has exhibited significant weight reducing property in various phase-3 SUSTAIN and PIONEER trials over dulaglutide and liraglutide. These results encouraged the researchers to carry out trials on weekly-once subcutaneous semaglutide for better management of obesity.

A cross over trial study on 30 obese subjects investigated the mechanism of semaglutide by studying energy intake, appetite sensation, gastric emptying, and postprandial glucose as well as triglyceride metabolism. Participants received weekly-once s.c. semaglutide (initially 0.25, then 0.5 and finally 1.0 mg with dose escalation) or placebo for 12-weeks, followed by a washout period of 5 to 7 weeks, and finally another 12-weeks treatment period. Semaglutide has shown about 24% drop in net calorie consumption with 5 kg weight reduction. Therefore, lower energy intake, enhanced satiation, less food craving, reduced body fat and weight loss are the plausible mechanism for the anorexic effect of semaglutide.

Semaglutide is a clinically approved drug for type 2 diabetes, obesity, and a clinical trial candidate for management of NASH, Parkinson’s and Alzheimer’s disease. Semaglutide is well tolerated and can be safely used in patients with renal, hepatic and CV diseases. Hepatoprotective and neuroprotective role of semaglutide in abating NASH and neurodegenerative diseases respectively has been proved in phase-2 trials and is still under study. It improves insulin resistance in the periphery as well as in the brain, reduces body weight, and reduces fat content in the liver by acting as an agonist on GLP-1 receptors. The efficacy of semaglutide in alleviating cardiac injury in COVID-19 patients with diabetes, obesity, hypertension and CV diseases is being studied. Based on drug repurposing strategy, researchers are trying their best to explore various therapeutic potentials of semaglutide. Clinical efficacy in different therapeutic fields proved semaglutide to be an indispensable treatment option for better management of “diabesity” and an emerging cynosure for management of NASH and neurodegenerative diseases.
